Conformational Transitions of Double-Stranded DNA in Thin Films

Conformational transitions of double-stranded DNA in different environments have long been studied as vital parts both vitro and vivo processes. In this study, utilizing Fourier transform infrared spectroscopy (FTIR), we provide detailed analysis dynamics A- to B-form thin films hydrated states based on a statistical substantial number spectra band shape (peak fitting) the phosphate (1150–1000 cm?1) sugar–phosphate (900–750 region. Hydration is systematically controlled by time spent desiccator chamber (from 3 min 40 min) allowing conformation hydration signatures, addition variations due ambient conditions, be resolved spectra. Conformation transition from A-form more ordered observed if sufficient allowed confirmed changes bands at ?890, 860, 837, 805 cm?1. Phosphate vibrations ?1230 cm?1 1089 cm?1, backbone ?1030 765 were found sensitive rather than conformation. Additionally, that spectral caused conditions can significantly reduced without inducing conformational changes, which serves good basis for quality assurance.
